Early Tests of an Appetite Hormone Shot and Combo to Treat Obesity

Ascletis, a biotech company, has started early-stage (Phase I) human studies of a new weight-loss drug called ASC36 and a combined version that pairs ASC36 with a GLP-1/GIP therapy. In plain terms, they’ve moved these experimental medicines from the lab into first tests in people to check safety, how the body handles them, and initial signs of effect. This is the very first step in a long process before any drug could be approved. ASC36 is described as an amylin agonist. Amylin is a natural hormone made by the pancreas that helps control appetite and the speed at which the stomach empties after eating. Saying ASC36 is an “agonist” means it’s a molecule designed to mimic amylin’s action — essentially pretending to be that hormone so the body gets the same signals: feeling fuller and slowing digestion. The other piece in the co-formulation targets GLP-1 and GIP, which are gut hormones that also reduce appetite and affect blood sugar. Combining these types of hormones is a strategy companies use to try to boost weight-loss effects. The news is about Phase I trials, which are small, early studies typically done in healthy volunteers or a limited number of patients. These studies mainly look at safety, dosing, and how the drug behaves in the body (for example, how long it lasts). The announcement doesn’t report outcomes like how much weight people lost, because Phase I is not designed to prove effectiveness. It also doesn’t say how many people are enrolled or provide results yet; it simply confirms the trials have begun. Why this matters is that obesity is a widespread health challenge and current medications, like semaglutide (the active ingredient in Ozempic and Wegovy), have shown significant weight loss in many people. New approaches that use amylin plus GLP-1/GIP signals could offer another option, possibly improving effectiveness or tolerability for some patients. If these early tests go well, the drugs would still need more extensive testing to show they work and are safe for larger groups before doctors could prescribe them. There are important caveats. Phase I trials don’t prove a drug works, and many candidates that look promising early fail later due to safety problems or lack of benefit. Short-term side effects for drugs in this class can include nausea, vomiting, and digestive issues; long-term risks are often less well known until bigger trials run for longer. Regulatory approval is still far off; starting Phase I is only the first of several required steps. Also, the announcement doesn’t provide details like participant numbers, trial design, or timing, so uncertainty remains about how quickly this could progress. Bottom line: Ascletis has begun first-in-human testing of an amylin-based drug and a combined amylin/GLP-1/GIP formulation for obesity, but these are very early safety-focused studies and it will take years of trials to know whether they’re effective and safe for general use.
