A Growth-Hormone Shot Made Nights Worse: New Wrist Numbness, User Reports

A reader started a drug called tesamorelin about six weeks ago and says they felt better within days and saw more changes by week three. They were already fairly fit and had their diet under control. Around week four they began getting moderate to severe carpal tunnel–type symptoms at night, waking several times. They’ve heard numbness or tingling is a known side effect and want to know why it happens and whether to keep taking it. Tesamorelin is a synthetic (man-made) peptide — a short chain of amino acids — that acts like a hormone-stimulating signal. It doesn’t act directly on tissues the way insulin does. Instead it tells the pituitary gland in the brain to release more growth hormone–releasing hormone, which then raises levels of growth hormone and a related factor called IGF-1. In medical practice, tesamorelin is approved for reducing excess belly fat in people living with HIV. People sometimes use it off-label for other reasons because it changes metabolism and body composition. The report here is a single-person experience, not a controlled study. Clinical trials and drug information do list peripheral neuropathy-type symptoms, including numbness, tingling, or carpal tunnel–like problems, as reported side effects for tesamorelin and other growth-hormone–raising treatments. The timing matches what’s described: neuromuscular symptoms can show up after weeks of use. However, we can’t quantify risk or how common or severe it will be from one anecdote. In large trials the majority of patients don’t get severe nerve problems, but a noticeable minority report sensory symptoms.
